Analysis of pain markers and epidural fibrosis caused by repeated spinal surgery in Sprague–Dawley rats

Abstract Background Epidural fibrosis is one of the aetiologies pain following a spinal revision surgery. It reported that specific members mitogen – activated protein kinases (MAPK) family might mediate neuropathic pain. However, roles epidural caused by repeated surgeries and pain-related proteins in causing post surgery syndrome remain unknown. Using rat adhesion model, this study, we evaluated investigated relationship between markers fibrosis. Methods Sprague–Dawley rats underwent were divided into three groups: group A (single laminectomy), B (two surgeries) C (three surgeries). Dural thickness was measured each experimental group, immunohistochemical analysis western blotting mitogen-activated performed (ERK, p38 JNK) using spine cord. Results 6.363 ± 1.911 μm A, 13.238 2.123 19.4 2.115 C, respectively. In blotting, phosphorylated ERK expression gradually increased with number surgeries, groups (1.77-fold) (2.42-fold) as compared to A. Phosphorylated showed an increasing trend (1.17-fold) (1.33-fold) JNK did not increase (1.62-fold) (1.43-fold) Excluding JNK, revealed dorsal horn, evidenced blotting. Conclusions Repeated may dural it suggests likely induced increases surgeries.
